A tecnologia Ion Torrent baseia-se na dete\u00e7\u00e3o direta atrav\u00e9s de um chip semicondutor, o que se traduz num fluxo de trabalho \u00e1gil, robusto e especialmente pr\u00e1tico para laborat\u00f3rios de investiga\u00e7\u00e3o e investiga\u00e7\u00e3o cl\u00ednica.<\/p>\n\n\n\n<p>A Ion Torrent realiza a sequencia\u00e7\u00e3o detetando as altera\u00e7\u00f5es de pH que ocorrem em cada po\u00e7o do chip quando a polimerase incorpora um nucle\u00f3tido e liberta um i\u00e3o hidrog\u00e9nio (H\u207a). Esta altera\u00e7\u00e3o de pH \u00e9 medida por sensores i\u00f3nicos e convertida num sinal el\u00e9trico, permitindo uma dete\u00e7\u00e3o direta, sem c\u00e2maras, scanners ou sistemas \u00f3ticos. A incorpora\u00e7\u00e3o de nucle\u00f3tidos \u00e9 registada em segundos e o processo ocorre em paralelo em milh\u00f5es de po\u00e7os.<\/p>\n\n\n\n<p>Os sistemas Ion GeneStudio\u2122 S5 foram concebidos para uma sequencia\u00e7\u00e3o dirigida flex\u00edvel e escal\u00e1vel, permitindo passar \u2014 dentro do mesmo fluxo de trabalho \u2014 de projetos pequenos para projetos de maior dimens\u00e3o sem necessidade de alterar a plataforma, bastando selecionar um chip diferente. Est\u00e3o dispon\u00edveis cinco op\u00e7\u00f5es de chip, oferecendo um throughput desde 2\u20133 milh\u00f5es de leituras at\u00e9 100\u2013130 milh\u00f5es de leituras. Isto permite otimizar o custo por amostra e responder de forma \u00e1gil a diferentes necessidades (desde pain\u00e9is pequenos at\u00e9 pain\u00e9is mais amplos), mantendo o mesmo fluxo de trabalho.<\/p>\n\n\n\n<p><\/p>\n\n\n\n<h4 class=\"c-title-4 wp-block-heading\">Automatiza\u00e7\u00e3o e an\u00e1lise integrada<\/h4>\n\n\n\n<p>O Ion Chef\u2122 automatiza a prepara\u00e7\u00e3o da matriz e o carregamento do chip com consum\u00edveis em formato de cartucho, minimizando a interven\u00e7\u00e3o do utilizador. A rastreabilidade completa das amostras e reagentes \u00e9 garantida atrav\u00e9s da integra\u00e7\u00e3o em rede com o software Torrent Suite\u2122.<\/p>\n\n\n\n<p>O Ion Reporter\u2122 Software automatiza a an\u00e1lise bioinform\u00e1tica, integrando, anotando e interpretando as variantes identificadas. Ao selecionar o workflow de an\u00e1lise adequado, a an\u00e1lise pode ser iniciada automaticamente ap\u00f3s a sequencia\u00e7\u00e3o, acelerando a passagem de dados para resultados.<\/p>\n\n\n\n<p>Gra\u00e7as \u00e0 tecnologia Ion AmpliSeq\u2122 utilizada na prepara\u00e7\u00e3o de bibliotecas, os instrumentos Ion GeneStudio\u2122 S5 podem trabalhar com quantidades muito reduzidas de ADN ou ARN (at\u00e9 1 ng), mesmo em amostras complexas como FFPE ou cfDNA. Em conjunto com o amplo portef\u00f3lio de pain\u00e9is Oncomine\u2122 validados e workflows espec\u00edficos para tumor s\u00f3lido e onco-hematologia, esta tecnologia \u00e9 particularmente adequada para laborat\u00f3rios de anatomia patol\u00f3gica e hematologia.<\/p>\n\n\n\n<p><\/p>\n\n\n\n<h4 class=\"c-title-4 wp-block-heading\">Aplica\u00e7\u00f5es em sa\u00fade reprodutiva e investiga\u00e7\u00e3o personalizada<\/h4>\n\n\n\n<p>Para laborat\u00f3rios de sa\u00fade reprodutiva e investiga\u00e7\u00e3o translacional, o Ion ReproSeq\u2122 (PGS\/PGT-A, RUO) permite a an\u00e1lise de aneuploidias a partir de amostras com quantidades muito reduzidas (por exemplo, bi\u00f3psia embrion\u00e1ria). Com o Ion GeneStudio\u2122 S5, o laborat\u00f3rio pode ajustar o rendimento da corrida utilizando diferentes chips (510\/520\/530), otimizando tempo e custo de acordo com o volume de amostras. A automatiza\u00e7\u00e3o com o Ion Chef\u2122 simplifica a prepara\u00e7\u00e3o e o carregamento do chip, reduzindo a manipula\u00e7\u00e3o e a variabilidade. Com o Ion AmpliSeq\u2122 Designer \u00e9 poss\u00edvel criar pain\u00e9is personalizados ao n\u00edvel de amplic\u00e3o: podem ser desenhados com base em genomas de refer\u00eancia pr\u00e9-carregados ou para qualquer organismo, carregando a refer\u00eancia em formato FASTA.
